Baric ผู้เชี่ยวชาญด้านโคโรนาไวรัสแห่งมหาวิทยาลัย North Carolina โดยค้นคว้าวิจัยความสามารถของไวรัสจากค้างคาวที่จะนำโรคมาสู่คน Dr.Baric ได้ถ่ายทอดความรู้ในการตัดต่อพันธุกรรมของโคโรนาไวรัส ทำให้สามารถไปติดเชื้อในสัตว์สปีชีส์อื่นๆ ให้แก่ Dr.Shi โดยทำการทดลองในหนูตัดต่อพันธุกรรมให้มีการแสดงออกของตัวรับ ACE2 receptor ของมนุษย์

เมื่อ Dr.Shi กลับมายังอู่ฮั่นก็ได้พัฒนางานวิจัยนี้ต่อโดยใช้เซลล์จากมนุษย์แทนหนู (มีหลักฐานของงานวิจัยนี้ที่ NIH เพราะได้ทุนจาก NIAID ผู้ลงนามรับรองคือ Dr.Daszak แห่ง EcoHealth Alliance)

เมื่อ พ.ย.2015 ทีมวิจัยนี้สามารถสร้างไวรัสชนิดใหม่โดยนำโครงสร้างหลักจากไวรัสซาร์ส SARS1 virus แล้วเสียบด้วย spike protein จากไวรัสค้างคาวชนิด SHC014–CoV พบว่าไวรัสตัดต่อพันธุกรรมนี้สามารถที่จะติดเชื้อได้ในเซลล์จากทางเดินหายใจของมนุษย์ สายพันธุ์ไวรัสตัดต่อพันธุกรรมใหม่นี้ชื่อว่า SHC014–CoV/SARS1 เป็นลูกผสม (chimera) ที่เกิดจากพันธุกรรมของไวรัส 2 สายพันธุ์

9 ธ.ค.2019 ก่อนการระบาดของโควิดไวรัส Dr.Daszak ได้ให้สัมภาษณ์ว่าขณะนี้นักวิจัยของสถาบันไวรัสวิทยาในอู่ฮั่นสามารถตัดต่อโปรตีนส่วน spike (S) protein และสร้างไวรัสโคโรนาลูกผสมที่สามารถติดเชื้อในหนูที่ตัดต่อพันธุกรรมของมนุษย์ และกล่าวในบทสัมภาษณ์ว่าใน หนูทดลองเหล่านี้จะป่วยเป็นโรค SARS ที่ไม่สามารถรักษาด้วย monoclonal antibody และไม่สามารถป้องกันด้วยวัคซีน


ผู้สัมภาษณ์ถามไปว่า "อ้าวในเมื่อรักษาไม่ได้ ฉีดวัคซีนป้องกันก็ไม่ได้ แล้วจะทำไงต่อ" (คงชักกลัว) Dr.Daszak ตอบ "ไม่ต้องกังวล ทุกอย่างสามารถควบคุมได้ไม่ยากในห้องแล็บ และงานวิจัยแบบนี้มีคุณค่ามากกว่าเยอะเพื่อจะลงลึกถึง spike protein ที่นำมาใช้สร้างวัคซีนในอนาคต"

ประเด็นหนึ่งที่อาจเป็นตัวต่อที่สำคัญของจิ๊กซอว์ปริศนาที่มาของโควิดไวรัส ก็คือ spike protein ที่มันใช้ในการจับและเข้าเซลล์มนุษย์ spike protein ของไวรัสจะมีอยู่ 2 หน่วยย่อยที่มีหน้าที่ต่างกัน คือ S1 ทำหน้าที่จับกับเป้าหมายของไวรัส คือ ACE2 บนผิวเซลล์ระบบทางเดินหายใจของมนุษย์ และ S2 ทำหน้าที่เชื่อมกับผนังเซลล์และช่วยให้ไวรัสสามารถเข้าเซลล์ได้

ไวรัสจะสามารถเข้าเซลล์ได้ก็ต้องตัดส่วน S1 และ S2 ออกจากกันก่อน ตำแหน่งที่ตัดนี้เรียกว่า furin cleavage site อยู่ตรง S1/S2 junction โดย furin ที่ใช้ตัดนี้เป็น protease enzyme อยู่ที่ผิวเซลล์ของคนเรา มันจะตัด amino acid sequence ที่เป็น proline-arginine-arginine-alanine-arginine (PRRAR) ซึ่ง sequence นี้อยู่บน furin cleavage site ของโควิดไวรัส ซึ่งเป็นตำแหน่งเล็กๆของไวรัสแต่มีบทบาทสูงต่อความสามารถในการติดเชื้อ

ปริศนาคือ มีแค่เจ้าโควิด SARS2 หรือ SARS-CoV-2 นี้เท่านั้นที่มี furin cleavage site (FCS) อยู่ตรง S1/S2 junction แถมยังเป็น loop ที่ยาวทำให้ furin protease มาตัดได้ง่าย ในขณะที่ SARS-related .a-coronavirus ส่วนใหญ่จะตัด S2 subunit ที่ตำแหน่งต่างออกไป

ดังนั้น จึงเกิดคำถามว่า FCS ตรงตำแหน่งนี้เกิดขึ้นเองตามธรรมชาติ (ซึ่งก็เป็นไปได้) หรือว่าจงใจทำให้เกิดขึ้นเพื่อการทดลองที่เรียกว่า gain–of–function experiment ที่ทำให้ไวรัสเก่งขึ้น ขอสรุปคร่าวๆเลยนะครับว่าเมื่อพิจารณาถึงประเด็นจากธรรมชาติแล้วพบว่ามีโอกาสเป็นไปได้น้อยมากทั้งจาก mutation และ recombination (ก็สมาชิกใกล้เคียงของตระกูลนี้ไม่มีใครเป็นแบบนี้เลย) ดังนั้น ก็มาถึงประเด็นที่สองคือผลิตขึ้นในห้องทดลอง (gain-of-function experiment)

gain-of-function experiment คือการทดลองเกี่ยวกับเชื้อโรคที่มีจุดมุ่งหมายในการชักนำให้เกิดการกลายพันธุ์เพื่อเพิ่มความสามารถในการติดเชื้อและการทำให้เกิดโรค รวมถึงการขยายศักยภาพของเชื้อให้สามารถติดเชื้อได้ในสปีชีส์อื่น โดยมีจุดมุ่งหมายเพื่อรับมือกับโรคติดเชื้อใหม่ๆ เพื่อพัฒนาวัคซีนและการรักษาโรคติดเชื้อ
